Location for cell viability analysis in real-time works on the basis of impedance monitoring in the area of electrodes which are placed on the bottom of the vessel to which the live cells are adhered. Impedance means electrical resistance for alternating current. Removable low voltage is less than 20 mV, resulting in an electric field between the electrodes, which interacts with the ionic environment of the growth medium present in the wells. The impedance is measured in cells that adhere to the bottom of the wells, so this method is only applicable to cells that are able to cling to these surfaces. If more cells sit on the surface of electrodes placed at the bottom of the wells, the value of electrical impedance will be higher. The resulting impedance is then expressed as CI (cell index). Instrument xCELLigence RTCA DP (dual plate) is able to analyze a larger number of plates simultaneously. The machine is further composed of four basic units: RTCA analyzer station RTCA, RTCA control unit, E-plate. Glass bottom of holes of measuring plates are coated with capillary gold electrodes.
